Williamsville was originally settled by the Quakers in the 1800s who developed the land into residential and commercial spaces for their members. They established the first lending library, school, and major road systems leading from Williamsville to Buffalo. In 1851, German immigrants settled land previously belonging to the Seneca Native American tribe and expanded the boundaries of Williamsville. Through the centuries, the neighborhood grew to become a large farming community with historical architecture - several homes and buildings are listed on the National Register of Historic Places.

Glen Falls Art Festival has been an Williamsville tradition since 1961. Artists and crafters come from all over the United States to celebrate their craft with other artists, residents, and tourists in Williamsville. The event hosts over 200 vendors and features food trucks from popular restaurants in Orchard Bark and Buffalo.
